(一)本组简介
本组主要负责人为程通教授,成员目前有助理教授3名、工程师1名、博士后1名、博士研究生7名、硕士研究生12名和实验技术员6名,是一个富有创新精神和凝聚力的科研小组。主要研究方向包括重要新发/突发传染病的广谱中和表位和新型疫苗、病毒感染和致病机制、新型溶瘤病毒、人源化动物模型和抗衰老功效因子等研究。近5年内,本组承担了重大传染病防治国家科技重大专项课题、重大新药创制国家科技重大专项课题、国家自然科学基金等多项课题研究,在Cell Host & Microbe、Nature Microbiology、Science Advances、Nature Communications、Gut、PNAS、Small methods、Cell Reports、Cellular & Molecular Immunology、EMI、Antiviral Res等权威学术期刊发表研究论文60余篇,已获得12项中国发明专利授权和6项美国、欧盟、日本发明专利授权。
本组组近年来取得了多项重要研究成果:(1)与美国Rutgers大学合作,应用病毒功能基因组学与反向遗传学技术研制出新型水痘病毒活疫苗(VZV-7D),是世界首个皮肤与神经双减毒水痘活疫苗,获得国家一类新药临床试验批件,目前已进入II期临床试验研究;(2)在儿童手足口病(HFMD)病原体肠道病毒的优势中和表位发现和新型多价疫苗研究上取得突破,应用分子病毒学与结构生物学技术发现了重要肠道病毒病原体CVA16、CVA6、CVA10、EV-D68和CVB1的免疫优势和广谱中和表位的精确结构特征与抗病毒作用机制,发现了广谱治疗性抗体和主要保护性免疫原,指导新型多价疫苗和特异性治疗药物研究,相关成果分别发表于Cell Host & Microbe、Nature Microbiology、Science Advances等权威期刊;(3)与浙江大学合作,建立了基于人干细胞移植的人肝细胞和人免疫细胞双重嵌合的人源化小鼠模型,可高度模拟人乙肝病毒的自然感染和乙肝肝硬化发病过程,相关成果发表在国际权威胃肠肝病学杂志Gut;(4)建立了高效的可支持新冠疫苗免疫原性评价、治疗性抗体和抗病毒药物筛选的体外评价模型,合作建立了新冠病毒的地鼠感染模型,为成功研制新冠诊断试剂和新型疫苗提供了重要支持;(5)开展了对胰腺癌、肝癌、肺癌等具有较好体内外溶瘤潜能的新型溶瘤病毒研究,在新型植物病毒载体和抗衰老功效因子研究等方面取得重要进展。
