人员构成:教师3人、高级工程师1人、博士后2人、学生45人、技术员7人,共58人。
研究方向:基因工程疫苗研究与开发、重要传染病病毒免疫表位的结构和功能研究、基于结构的分子模拟和分子设计、生物大分子药物质量分析与制剂研究、新型疫苗佐剂研发。
技术平台:基因工程疫苗研发平台、基于CRISPR-Cas9技术的大肠杆菌基因组改造、杆状病毒表达系统、哺乳动物细胞表达系统、X-ray衍射晶体结构解析平台、Cryo-EM结构解析平台、分子动力学计算平台、二代测序平台、生物大分子药物质量分析平台、佐剂研发平台。
研究内容:
1. HPV系列:基于结构的人乳头瘤病毒(HPV)型交叉疫苗研发、HPV抗体中和机制研究、基于HPV VLP的载体展示/递送平台、HPV次要衣壳蛋白、晚期蛋白结构和功能研究、HPV感染关键因子筛选和相关感染机制研究。
2. HEV系列:戊型肝炎病毒(HEV)广谱中和抗体和型特异抗体表位及机制研究、基于HEV结构设计的通用的颗粒化疫苗研究、基于体外偶联技术和结构设计的多糖联合疫苗研发。
3. VZV系列:基于水痘-带状疱疹病毒表面糖蛋白gE的重组疫苗研发与结构解析研究。
4. HIV系列:基于HIV-1包膜蛋白的免疫原设计及抗原免疫原性、广谱性的探索;HIV-1全长膜蛋白的提取及结构研究。
5. 大肠杆菌表达系统系列:基于gp2基因的大肠杆菌底盘细胞改造和应用、戊肝P239颗粒纳米抗体原核表达系统构建。
6. 肠道病毒、轮状病毒、新冠病毒、呼吸道合胞病毒、猴痘病毒等重要病原体的结构研究:解析肠道病毒多种衣壳形式和轮状病毒、以及新冠病毒、呼吸道合胞病毒膜蛋白与中和抗体复合结构,研究抗体介导的病毒中和作用机制、以及基于结构的中和抗体的广谱作用研究。
7. 疫苗质量控制:重组疫苗相关质量分析方法的建立(生物物理、生物化学、免疫化学方法)、重组疫苗体外相对效力分析、生物药物后期转化。
8. 新型佐剂:基于递送载体的佐剂递送系统研发;基于结构设计靶向型佐剂分子研究;基于有机无机杂合纳米佐剂研发;佐剂免疫调节机制研究;佐剂肿瘤治疗与预防机制研究。
